Further information and research on Temozolomide

Review of Temodar (temozolomide) - Written in 2000 on www.virtualtrials.com by Andrew Parker, Dept. Biomedical Science, University of Sheffield. This report lists many of the recently published articles on temodar ( Temozolomide) and attempts to set them into the context of the development of temodar (temozolomide).

Journal articles and abstracts on Temozolomide - A list of links on www.virtualtrials.com to journal articles and abstracts on temozolomide. Virtual Trials introduction to this sections says "Since our excellent review of Temodar by Andrew Parker was completed in June, 2000, there have been many journal articles and abstracts presented on Temodar. We will list a brief synopsis, with a link to the reference (or abstract if available) of the most important ones here. Check back for updates. The most recently added ones will be at the top!"

Temozolomide - Standard of care (5/23 schedule):

  1. Radiotherapy plus concomitant and adjuvent Temozolomide for newly diagnosed Glioblastoma PDF document. Adobe Acrobat required. - New England Journal of Medicine article on a trial comparing Radiotherapy alone and radiotherapy plus temozolomide, given concomitantly with and after radiotherapy, in terms of efficacy and safety.
  2. "For a retrospective analysis which suggests that TMZ should be continued as long as it remains effective in controlling Tumor growth (rather than continuing only 6 months after completing radiotherapy, per the Stupp et al protocol), see Anderson and Lindsey 2008 PDF document. Adobe Acrobat required.; however, note that this analysis has at least two methodological problems: (a) patients with both Grade III and IV gliomas are aggregated together, and (b) only patients who took TMZ for at least 12 months were included in the analysis, thus biasing the results by excluding patients who passed away before 12 months." - Source: www.brain-treatments.net
  3. Balmaceda et al 2008 - Multi-institutional phase II study of temozolomide administered twice daily in the treatment of recurrent high-grade gliomas.

Temozolomide on a daily schedule:

  1. Buttolo et al 2006 - Journal of clinical oncology article on alternative schedules of adjuvant temozolomide in Glioblastoma Multiforme: A 6-year experience.
  2. Kurzen et al 2003 PDF document. Adobe Acrobat required. - Inhibition of angiogenesis by non-toxic doses of temozolomide.
  3. Baruschel et al 2006 PDF document. Adobe Acrobat required. - Safety and pharmacokinetics of temozolomide using a dose-escalation, metronomic schedule in recurrent paediatric brain tumours.
  4. Kim et al 2006 - Metronomic treatment of temozolomide inhibits tumor cell growth through reduction of angiogenesis and augmentation of apoptosis in orthotopic models of gliomas.
  5. Kong et al 2006 - A pilot study of metronomic temozolomide treatment in patients with recurrent temozolomide-refractory glioblastoma.
  6. Zhou et al 2007 PDF document. Adobe Acrobat required. - Preclinical pharmacokinetic and pharmacodynamic evaluation of metronomic and conventional Temozolomide dosing regimens.
  7. Perry et al 2008 - American society of clinical oncology article on the temozolomide RESCUE study: A phase II trial of continuous (28/28) dose-intense temozolomide (TMZ) after progression on conventional 5/28 day TMZ in patients with recurrent Malignant Glioma.
